Quantitative Assessment of Periodontal Bacteria Using a Cell-Based Immunoassay with Functionalized Quartz Crystal Microbalance

Periodontal disease is an inflammatory disorder that triggered by bacterial plaque and causes the destruction of tooth-supporting tissues leading to tooth loss. Several bacteria species, including Porphyromonas gingivalis Aggregatibacter actinomycetemcomitans, are considered be associated with severe periodontal conditions. In this study, we demonstrated a quartz crystal microbalance (QCM) immunoassay for quantitative assessment bacteria, A. actinomycetemcomitans. An immunosensor was constructed using self-assembled monolayer 11-mercaptoundecanoic acid (11-MUA) on gold surface QCM chip. The 11-MUA layer evaluated cyclic voltammetry technique determine its mass packing density. Next, monoclonal antibody covalently linked 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide/N-hydroxysuccinimide act as biorecognition element. specificity confirmed enzyme-linked immunosorbent assay. A calibration curve, relationship between frequency shifts number used calculate actinomycetemcomitans in test sample. Based regression equation, lower detection limit 800 cells, dynamic range up 2.32 × 106 cells. Thus, biosensor study provides sensitive label-free method analysis bacteria. can various biosensing assays practical application routine periodontitis pathogens.
